Cellular health and organismal longevity are dependent upon the Proteostasis Network (PN) of molecular chaperones, transport pathways and clearance machineries that regulate the synthesis, folding, and function of the proteome. Robustness and optimization of the PN by cell stress responses that respond to environmental dynamics is critical to aging and longevity, with failure in proteostasis leading to neurodegeneration and other diseases associated with misfolding and aggregation. I will discuss how the functional properties of the PN prevent the toxicity of highly aggregation-prone proteins in neurodegenerative disease, evidence that PN failure in aging releases a cascade of misfolding and aggregation of other metastable proteins, and that transcellular communication between neurons and surrounding tissues can reactivate the heat shock response to restore cellular and tissue health and longevity.

Really cool explanation. A lot of proteins naturally aggregate and function as biomolecular condensates, but these aggregates are tightly regulated from becoming harmful by the proteostasis network (chaperones (i.e. refolding proteins), degradation, sequestration). Proteostasis failure causes disease.
