To me it felt more like Shinobu was angry because she was thinking about the demon who killed her sister. She was going to tell Kanao the story how Kanae was killed.

Shinobu doesn't hate Nezuko, but she didn't trust tamayo at first because Tamayo was a demon, but later on, she actually praised Tamayo say that she was nice!

Personally, I think the part when she's angry in front of Kanae's altar is an accumulation of many things. We are humans after all, sometimes our anger comes from various reasons. For Shinobu's case, I think its a combination of unable to participate in the training, getting reminded of the demon that killed Kanae, and needing to work alongside with a demon (I've a feeling she has pride in her pharmaceutical skills and the creation of her poison, so having to work with a demon for that might've made her feel a bit insulted). All in all, Shinobu is a complex and multilayered character that I really like a lot.

Also, while their relationship wasn't explored much in the series, Nezuko and Shinobu had a decent relationship where it was shown from Nezuko's memories, Shinobu does treat her nicely.

Actually I think the reason why she was mad was because she remembered douma the demon who kill her sister because after that she spoke with kanao about it directly so this is the main reason

The only reason she’s probably comfortable with Nezuko is since she got to know her better and/or knows she’s a good demon. On the other hand, She had just met Tamayo and Yushiro, so she had a valid reason to be uncomfortable with them, or feel like Yushiro was being a bit to excessive / overprotective of Tamayo, which caused her uncomfortableness.
