I always knew Great Unclean Ones to be fairly cheerful demons, humming a tune and actually giving a shit about their underlings, they see them as their children

So it just hits pretty hard to hear genuine anger with some bitterness and that tone of voice that can come only from someone who has been genuinely emotionally hurt

This is actually fairly lore-accurate. Many Great Unclean Ones believe only Nurgle himself should brew new plagues, and it's the duty of the Great Unclean Ones to spread Grandfather's gifts. So Ku'gath performing his own experiments is seen as somewhat heretical.
