Sig ability: Not necessary but makes a big difference in longer fights. Having high sig level also helps in throwing two heavies to get the massive poison damage twice during frenzy (attack increase you get after sp3).
Powerful Synergies: 1) Daredevil one is great. Extra 2 seconds of poison= 14k damage for my 5/65 Mysterio but running Daredevil isn't very practical.
2) Stealth Spidey: This is great. With this you can increases the duration of Frenzy and the illusion (power drain from sp2) for 2 seconds which is all you need most of the time to get that second mega poison off.
3)Emma Frost: Probably the most famous one. Makes him immune to reverse control which is something that is becoming more and more useful as the game progresses.
4)PENNY PARKER: Most imp synergy. Allows you to heavy without loosing charges
Sp1: Not very practical in endgame content. I showed an example of it working and failing in my video. With the massive attack ratings you can't really get hit and then reflect it reliably cos you will be dead 90% of the time. With the Heimdall synergy it can have some uses for sure but generally speaking mostly stick to sp2/3.
Power Control: Its really strong but you do sacrifice on damage. I wish that he worked like Symbiote Supreme and got all three abilities for a while after sp3s. That would not only make power control more useful but the damage reversal would also be kind of like a safety net like Magik's limbo.
Masteries: Suicide friendly unless you wanna power control. Poison immune and you would usually just spam sp3s and heavy attacks so no recoil damage.
