Clint Eastwood had played a few years before in Francis in the Navy, a movie about a talking mule. I think is that event in his past which inspired him to suggest to Leone the reason for the shootout, that the baddies had laughed at his mule and that the mule felt insulted by that,

The audio immediately takes me back to the Drive-In Movie Theater with it’s cheap, tinny-sounding speakers where I first saw ALL the Spaghetti Westerns 😂😃👍
