This lecture explains about the hotstart PCR mechanism and procedure. It also explains the hotstart PCR principle. Hotstart PCR is performed at high temperature at the starting of the PCR reaction to eliminate unwanted binding of primers. As primers are prevented to bind to the nonspecific region of the target DNA, unwanted product amplification is also prevented. So with the help of this hotstart PCR we can get more specific amplification of target DNA product.

how can polymerase activate in lower temperature....and how can polymerase bind while our DNA strructure is still double stranded before 95 degree?

I have a question please. In the 2nd step, or second way ( modification of primers). How these binding proteins can still normal after the 95C of the denaturation step, until arriving annealing step of 72C that will degrade it ?

Sir, after the binding proteins get denatured, the primers are free to form primer dimers, because unless the annealing temp comes they won't bind to the strands! Then how is it helping in preventing the primer diner formation! And moreover at 95 degree the binding proteins will denature, won't the primers denature too? Or are the thermostable like the polymerase?
