Welcome to Aladdin creations , My name is kavindu ….

In this video I’m going to discuss with you , how to differentiate immature white blood cell types …
Myeloblast , promyelocyte , myeloccyte , metamyelocytes & stab form or band Neutrophils …

So let’s begin this …

Let’s see how the maturation & cyto differentiation occur …
First primitive multipotential stem cells gives rise to , myeloblast … , then it becomes to promyelocyte , myelocyte , metamyelocyte , Band or stab form and finally mature in to granulocytes , such as Neutrophil , eusonophil or basophil.

So , how we can identify these cells in stained bone marrow smear …
For that we can use 6 major facts ,
• Nucleus shape
• Size of the cell
• Present of absent nucleoli
• Present of absent granules
• N/C
• Color of cytoplasm

Now we are going to see morphological features of each cell , in detail …

Mayeloblast

The first commited stem cell of granulocyte is myeloblast .
Usually it is spherical & about 10 – 18 um in size.
Nuclear to cytoplasm ratio is very high.
It has evenly dispursed chromatin with 2- 4 nucleoli .
And It does not contain granules .
However small cytoplasm is blue in color , due to high RNA content.

Promyelocyte

When we consider promyelocyte ,
It is about 12-18 um in size & usually little larger than myeloblast cells.
And actually these promyelocytes cells are the laregest immature WBC types.
Nuclear to cytoplasm ratio is high.
And the chromatin is slightly condensed .
Also similarly it has 2 – 4 nucleoli.
It contained reddish – purple scanty large granules
In faint blue color cytoplasm.

Myelocyte

When we talk about myelocyte ,
It is avoid in shape & little smaller than myeloblast.
It contain condensed chromatin
Also N/C ratio is low
And Remember we can not see nucleoli.
And other distinguishable feature is presence of many small granules.
Cytoplasm is faint blue to pinkish in color.

Metamyelocyte

When we consider metamyelocyte .
It is easy to identify because it has kidney shape nucleus which take half of space from it cytoplasm.
Also N/C ratio is low
It does not contain any nucleoli
But cytoplasm contain many primary & secondary granules
And Cytoplasm is pinkish color ???

Band / Stab form

Then metamyelocyes mature in to band or stab form.
The most specific feature of this cell is the presence of band shape neutrophil
Also N/C ratio is low
And it does not contain nucleoli
cytoplasm contain many primary & secondary granules
And Cytoplasm is pinkish color ???

When we consider the size of the cells , promylelocytes are the biggest cells ,myeloblast , mylocyte , metamyelocyte & stab forms or band nutriphills are the smallest cells …

Finally these cells give rise mature granulocytes …

Would've been a good video if you got a better mic and pronounced the words properly. Or atleast added subtitles. I can MAYBE make out every 5th word. Good illustrations tho.
