Culture Media (Part 1)- Animal Cell Culture

What is culture media ? It is a nutrient preparation that contains essential nutrients required by cells for their growth.
Two types of culture media :
- Natural media
- Artificial media
Natural media consists of naturally occurring biological fluids such as plasma clots, biological fluids like amniotic fluid, insect hemolymph, serum, tissue extracts.
Artificial media are grouped as:
- Basal media
- Serum media
- Serum free media
- Protein free media
Basal media - also termed as chemically defined media as all the components are known.
Components of Basal Media includes:
- balanced salt solution : provides an environment that maintains the structural and physiological integrity, pH and osmolarity of cells in vitro
- buffering system : regulates pH. these are of two types, natural and chemical
- energy source : commonly used is glucose. other sugars like maltose, sucrose and fructose are also used
- amino acids : increase cell proliferation.
- proteins/peptides : albumin act as binding protein, aprotonin act as protective agent and fibronectin aids in attachment of cells to the substrate
- growth factors : act as signal and helps in cell proliferation and differentiation
- vitamins : act as coenzymes involved in cell metabolism. eg vitamin B complex
- trace elements : copper, zinc and selenium are elements needed in micronutrients. These are essential for biological processes
- antibiotics : prevents contamination
- phenol red : serves as a pH indicator
