This video covers part one of the genetics section of the USMLE First Aid textbook. Basic terminology will be covered in this video:
Gene expression modifications:
- knock-in, and knock out (insert or remove a gene)
- random insertion: constitutive expression (a gene that is always on)
- targeted insertion: conditional expression (a gene that can be turned on or off at will)
Cre-lox system:
- turning genes off or on by an external stimulus to study gene at developmental points
RNA interference:
- miRNA (MicroRNA) - blocks mRNA translation leading to degradation
- siRNA (small interfering RNA) - an exogenous source that binds to specific mRNA leading to cleavage of mRNA before translation
Codominance:
- both alleles are expressed
Variable Expressivity
- can have the same genotype but different phenotypes
Incomplete penetrance
- not all mutants show the phenotype
Pleiotropy
- one gene makes multiple phenotypic effects
Anticipation
- increases the severity of the disease
Loss of Heterozygosity
- cancer requires mutation of the tumor suppressor gene AND complementary gene deletion/ mutation
