I just submitted a paper to Science, and have published there before. One thing you have to realize is that writing one of these manuscripts is very different than a standard publication. It requires really seeing the big picture and distilling your question and solution into a clear story. It also requires understanding that whether the editor thinks it belongs in Science or whether the reviewers like it enough is almost entirely unpredictable! It helps if you PI is famous though. ;)
