Just thought I'd list my five favorite parts of this video: 

1. "Hold on, let me blow this fly up real quick."
2. "This fly went to McDonald's every day of its life."

David Ebertz Hey, great video! but I'm just wondering, for when you want to express a gene locally in a certain area in an organism that doesnt have Gal4, like in Drosophila (in your example you used the eye), instead of producing 2 constructs, one to produce Gal4, and the second to use Gal4 to produce the protein...why can't you just make one construct, using the transcription factor found in the eye to transcribe the gene of interest? I don't see the requirement for Gal4 in that process? Cheers!

Gal4 binds to UAS, a promotor region that is in another strain of flies, that is mated with the Gal4 driver strain.  I'm not sure what you mean with your second construct?... Are you trying to compare tissue specific Gal4, or when Gal4 is expressed in all cells/tissue?
