I've always said, I think Quill knows he's from earth, but he's also really good at thinking on his feet. We also see that the aliens in guardians of the galaxy are extremely ignorant of earth (to be fair most of them never even heard of it). So him saying "I'm not from earth i'm from missouri" was a quick on the fly way for him to tell if the people he was talking to were really from earth. If they were thanos' crew they would've likely said something like "what is a missouri?"

Well, the better part of this scene is that Quill doesn't know that the others are from Earth because there are other planets where people look human, just like those on Xandar. This also explains why they came in fighting thinking it was Thanos's people they were attacking.

He hasn’t forgotten. If he remembers earth music, style, tv characters and remembers that him mother was from earth I doubt he’d forget the name of the planet.
