Canonically, Fry's brain couldn't be read in a previous episode due to him being his own grampa and, thus, not having enough delta brainwaves.
The Dark One is actually the last of a species that evolved from either frogs or snakes and, well, they're basically evil and want to wipe out all other life - it looks like a leech and has psychic powers. Their fated enemy is the Encyclopods, whom store different DNA, and the Dark One attempted to wipe them out by destroying the violet dwarf star, which Leela's feminist group attempted to stop at first.
