Acid-Fast Staining Procedure
Introduction:
Acid-fast staining is a microbiological technique used to differentiate acid-fast bacteria, such as Mycobacterium tuberculosis, from non-acid-fast bacteria. This staining method plays a crucial role in the diagnosis of diseases like tuberculosis.
Materials Required:
Smear slide with fixed bacterial specimen
Carbol Fuchsin solution
Acid-alcohol (decolorizing agent)
Methylene Blue or Brilliant Green (counterstain)
Bunsen burner or slide warmer
Microscope
Procedure:
Prepare a bacterial smear on a clean slide, ensuring the specimen is fixed using heat. This can be done by passing the slide briefly through a flame (Bunsen burner) or using a slide warmer.
Apply a few drops of Carbol Fuchsin solution to the smear, completely covering the specimen.
Heat the slide gently by passing it through the flame or using a slide warmer. Do not overheat, as it may cause excessive steam and loss of the stain.
Keep the slide warm for 5-10 minutes. Add more Carbol Fuchsin if it dries out during this step.
Rinse the slide with acid-alcohol (decolorizing agent) for about 15 seconds, ensuring the stain is removed.
Rinse the slide with water to stop the decolorization process.
Counterstain the smear with Methylene Blue or Brilliant Green for 1-2 minutes.
Rinse the slide with water to remove excess counterstain.
Blot the slide gently with a paper towel or bibulous paper to remove excess water.
Observe the stained smear under a microscope. Acid-fast bacteria will appear pink or red, while non-acid-fast bacteria will appear blue or green.
