If the blood smear was left to air dry overnight would that lead to a lack of romanowsky-effect, meaning less purple coloring and more a blue hue on the cell nuclei? I tried to optimize the process with the solution we had at hand, day 1 adjusting pH day 2 adjusting the time interval. I figured leaving the blood smears overnight wouldn't make a difference but they were overwelmingly stained with Eosin Y and there were NO purpur nuclei.. The only reason I can think of is the lack of hydrophobic stabilization, leading to no Eosin Y binding in the romanowsky effect
