Kinetic energy is calculated by 1/2mv^2. Because Projectile motion is 2D motion, at highest point of Projectile motion, vertical component of velocity is zero. Because vertical motion is affected by gravitational pull. At highest point, vertical velocity is zero. However, there still exists horizontal velocity. One of the assumptions we use in projectile motion is that uniform motion is constant. So whatever is the horizontal velocity in the start of the motion, it remains the same throughout the motion. So if initially, the horizontal velocity is Ux, at highest point of projection motion, it is still Ux. Hence Kinetic energy at highest point of projectile motion is 1/2m(Ux)^2.

Horizontal velocity being constant is not an assumption it is due to the fact that there is no acceleration in the x direction.
