Orexin, also referred to as hypocretin, is a neuropeptide that plays a crucial role in the regulation of arousal, wakefulness, and appetite. It is present in two distinct forms: orexin-A and orexin-B. The most prevalent variant of narcolepsy, known as type 1, is characterized by episodes of sudden muscle weakness, commonly referred to as "drop attacks" or cataplexy. This condition arises from a deficiency of orexin in the brain, resulting from the degeneration of the neurons responsible for its production.
In the human brain, there are approximately 50,000 to 80,000 neurons that produce orexin, primarily situated in the perifornical area and the lateral hypothalamus. These neurons have extensive projections throughout the central nervous system, influencing wakefulness, feeding behaviors, and various other functions. Orexin exists in two peptide forms and interacts with two distinct types of orexin receptors.
