The film definitely takes place between the Nun and Conjuring 2 takes place, like just before or after Frenchies exorcism. We know it takes place after The Nun as Sister Irene is wearing her Nun garments as in the trailer, as she took her vows in the Nun 1

Its similar to Conrad Veight in the 1928 film The Man Who Laughs, the origin of The Joker. The Nun actress (like Veight) just has one of those faces, just the right amount of peculiar bone structure to be a completely different person when painted up right.

I'm judging from the trailer I guess that this show is after the first nun movie and before the first conjuring movie .
My theory is that valak is still alive after the events of the second conjuring considering the fact that in the nun they vanish valak by giving him the blood of Jesus Christ but then returns so even with Loraine's chant they would not be able to completely destroy valak and will just return's from the near future.
