Viruses with DNA genomes must carry out DNA replication to produce nucleic acid for packaging into new virus particles. At least one protein, sometimes many, must firt be made for genome replication to take place and therefore this process is always delayed in infected cells. In this lecture we cover the mechanisms of DNA replication, including how origin-binding proteins recruit the host synthetic apparatus, replication by strand displacement or a replication fork, solving the 5'-end problem caused by RNA priming, how genomes of different structures are copied, and how viruses ensure that cells are actively duplicating their DNA to provide access to the DNA synthesis machinery.

I have a doubt or may be I feel the replication of the DNA is always primer dependent whereas in the rules it says may or may not be.

why at 31:43, one ITR can be nicked by DNA pol, while one needs nickase?
