Pain defeated both Tsunade and Orochimaru with just one Path of Pain, but Jiraiya was at least able to make him use all the Six Paths of Pain in order to defeated.

1. Orochirmaru (Boruto Era) he is a true immortal with instant regen due to white zetsu body composed of hashirama cells, has more knowledge and access to everything he could ever want.
2. Kashin Koji, perfected Clone of jirayah with everything jirayah had but better and more abilities.
3. Tsunade, huga chakra pool cus of the Senju/Uzumaki genes in her, incredible strength, the best summoning, beat orochirmaru (no hands) with ease, and has 100 healinfs so she can go near death 100 times and the byakugo seal is the only reason i out her above jirayah as jirayah would be forced try and kill someone like Tsunade about 100 times and evade every single punch as 1 shot would kill him.

Tsunade did not even fight pain since she was too busy healing and saving everyone in the village

Koji Kashin is the clone of Jiraiya and has all of his abilities and skills, and was almost able to kill a god( if Ishiki hadn’t come back to life, Jigen would have turned to hashes and Ishiki with him ).
Also, its obvious that Tsunade isn’t the strongest( i’m not saying she isn’t strong, i’m just saying that Jiraiya and Orochimaru are stronger ), so i’ll exclude her for now. Itachi encountered both Jiraiya and Orochimaru. He completely made fun of Orochimaru, cut an arm of him and forced him to leave the Akatsuki. On the other hand, when he encountered Jiraiya, he said that if he attacked him, he would ultimatly get slaughtered.Afterwards, Itachi fought against Jiraiya and had to run for his life if he didn’t want to slowly and painfully get melted. Not to mention that when he encountered Jiraiya, he was older than when he fought Orochimaru, wich means he was stronger and with more experience, and also had Kisame with him, who had been able to kill his six teammates( who weren’t just some random dudes, they were part of the Sordsman of the Mist, so the élite of élite in the Mist Village ) and beat the crap out of Might Guy( that guy who almost killed Madara Uchiha ).

Tsunade is physically the strongest, Orochimaru the smartest and overall Jiraya is better than both of them. Not to mention it was canon stated that Jiraya was the strongest of the Sannin and still would be had he not died.
