This is not the most damaging build for a blowgun, full fighter with sharpshooter would probably be the one. But it's more in flavor as blowguns are used to apply poisons, not pierce people's head.

Alternatively, you could fill your darts with healing potions (2D4+2, which would become 2D4+1 after subtracting the 1 damage of the blowdart) and be a distanced martial healer, who constantly gives his teammates steroid injections.

Pretty sure most GM's would let you use Animal Handling (or maybe Arcana given it's magic) to milk venom from a snake

Technically you can’t extract poison from a dead familiar since they just disappear when they reach 0hp

You could homebrew and have an artificer make poisons or potions to dip the darts in, or enchant the darts to have some other effect

You don't need to kill your snake for the venom. You just need to force it to release it with a jar.

Don't forget that milking a snake for their venom is a thing, so you don't even need to kill the snake familiar.

I can see an Air Genasi rocking this weapon. Since their whole thing is air, I imagine they'll be able to accelerate the dart to leathal effect.
